Fig. 8 Role of depletion of organic materials for the formation of electron microscopic image. Cultured fish scale melanophores were extracted with detergent and processed for electron microscopy. (a), Release of replica did not include organic material degradation; melanosomes (white oval structures) have high electron density, which does not allow the visualization of cytoskeletal fibers associated with pigment granules. (b), Release of replica included organic material degradation by Clorox Bleach; cytoskeletal fibers can be seen on the top of "footprints" of pigment granules. Bar, 0.5 µm.
